Cornish airline Skybus will start flights from Newquay to Cardiff International on March 10.
Tickets are now available at the introductory price of £35 one way if booked before January 31, 2008.
The service will run five days a week, from Monday through until Friday.
Jeff Marston, chief executive of the Isles of Scilly Steamship Company which operates the Skybus air service from Newquay, said: "We have been looking at expanding our service and entering new markets for some time. The Cardiff to Newquay route is the perfect development for our service.
"We know that the route has been really popular in the past, with another carrier managing the route during 2005 through until the start of 2007.
"We are delighted to be able to reinstate the route and offer a convenient, reliable and quick service for the people of Wales and Cornwall."
Skybus took delivery of a twin otter aircraft last month in order to service its route expansion programme.
